Most autosamplers use a six-way injection valve, which includes a circular seal (rotor) and a fixed base (stator). The stator is connected to the pipeline, sample ring, sample hole, and other peripheral components. It is generally made of wear-resistant materials such as stainless steel or ceramics. The rotor and stator are tightly combined and can be rotated during operation. They are generally made of softer materials than the stator. For example, the rotor of the autosampler is driven by an electric motor (some older models use compressed air to drive it), while the manual injector has a handle attached to the rotor.

The rotor includes some small grooves for connection to different stator positions. The rotor is in the load or fill position and the sample is filling the sample loop, and the pump directly sends the mobile phase into the chromatography column; the rotor is turned to the injection position, and the channel connected to the pump is connected to the sample loop, and the mobile phase flushes the sample into the chromatography column.
